In this experiment, GFP-labeled ovalbumin-specific CCR7-neg effector memory T cells with elevated Kv1.3 channel expression were adoptively transferred into the peritoneal cavity of healthy Lewis rats. Forty eight hour later, these rats were challenged with antigen (ovalbumin conjugated to Texas red) in one ear and with saline in the other ear. Two-photon imaging was used to visualize the green (GFP-labeled) effector memory T cells that had trafficked to the antigen-challenged ear of the rat 3 hours after antigen challenge. In this video, a green (GFP-labeled) effector memory T cell is seen making a contact with a red antigen presenting cell. The APCs appear red because they have taken up the antigen, ovalbumin-Texas red. An average contact is about 20 minutes.
