Effector memory T cells moving along collagen fibers in inflamed tissue 24 hours after antigen challenge during DTH

In this experiment, GFP-labeled ovalbumin-specific CCR7-neg effector memory T cells with elevated Kv1.3 channel expression were adoptively transferred into the peritoneal cavity of healthy Lewis rats. Forty eight hour later, these rats were challenged with antigen (ovalbumin conjugated to Texas red) in one ear and with saline in the other ear. The entry of the effector memory T cells (green cells) into the ear was visualized by two-photon microscopy. This video shows green effector memory T cells moving along collagen fibers (blue) in inflamed ear tissue of a Lewis rat 24 hours after challenge with antigen (ovalbumin conjugated to Texas Red). The antigen presenting cells are red due to uptake of the labeled antigen.
The effector memory T cells did traffic to the ear, but once within the tissue, the cells were immobilized. In this video, green effector memory T cells are stuck on the collagen matrix (blue) and are unable to interact with antigen presenting cells (red because they have taken up ovalbumin-Texas red).
